Vitrification, also known as flash-freezing, is a state-of-the-art cryopreservation technique used in reproductive medicine. Unlike conventional slow-freezing methods that can form damaging ice crystals within cells, vitrification eliminates ice crystal formation by rapidly transitioning the liquid state into a glass-like solid.
The Vitrification Process:
Embryos are treated with specialized cryoprotectant solutions.
They are instantly exposed to ultra-low temperatures.
Rapid freezing solidifies the cells without structural cellular damage.
Embryos are stored indefinitely in liquid nitrogen tanks at -196°C.
When needed, they are thawed and safely transferred to the uterus.
Benefits of Embryo Freezing:
Surplus high-quality embryos from a fresh IVF cycle are preserved for future use without discarding.
Patients can attempt subsequent pregnancies using frozen-thawed cycles with minimal medication.
Eliminates the risk of Ovarian Hyperstimulation Syndrome (OHSS) by postponing transfer in high-risk patients.
Sperm cryopreservation is the process of preserving sperm cells at sub-zero temperatures. It is recommended for men facing medical treatments (such as chemotherapy or radiation), individuals with declining sperm parameters, or cases where sperm is retrieved via surgical methods (micro-TESE/TESA).
Procedure Stages:
Sample Collection: Retrieved via masturbation or surgical sperm extraction.
Analysis and Processing: Evaluated for motility, concentration, and morphology, then washed and prepared.
Cryoprotectant Media: Blended with solutions designed to preserve cellular membrane integrity.
Freezing & Long-Term Storage: Cooled under controlled conditions and stored in liquid nitrogen for long-term viability.
Egg (Oocyte) Freezing
Oocyte cryopreservation allows women to freeze their eggs at a biologically younger age, preserving reproductive potential for medical reasons (oncological treatments, genetic predisposition) or personal/social factors.
Procedure Stages:
Ovarian Stimulation & Retrieval: Hormonal protocols stimulate follicle growth, followed by ultrasound-guided egg retrieval (OPU).
Assessment: The embryology laboratory isolates mature, healthy oocytes (MII).
Vitrification & Storage: Eggs are processed with cryoprotectants, instantly frozen, and stored securely in liquid nitrogen tanks.
ROSI Technique (Round Spermatid Injection)
ROSI is an advanced reproductive technique designed for severe male infertility cases (non-obstructive azoospermia) where mature spermatozoa cannot be found even through surgical exploration. It utilizes round spermatids—immature precursor sperm cells—which are injected directly into the mature egg using specialized micro-needles to achieve fertilization and blastocyst development.
Piezo-Electric Assisted Oocyte Activation (AOA)
Piezo-electric activation involves applying micro-electrical pulses to stimulate the egg following ICSI. It is utilized primarily for patients with previous total fertilization failure or extremely low fertilization rates.
Approximately 30 minutes after microinjection, eggs are placed between parallel electrodes in a specialized buffer solution inside an Electro Cell Manipulator. The controlled electrical pulse mimics natural physiological activation, triggering the signaling pathway required for the sperm and egg to fuse and complete fertilization. This technique significantly enhances embryo formation and clinical pregnancy rates in challenging cycles.
